Clinical Studies

Combined 0.1% retinaldehyde/ 6% glycolic acid cream in prophylaxis and treatment of acne scarring.

Products

Avène Diacnéal

Trial Design

Randomized, double-blind, vehicle-controlled, multi-center, parallel-group

Duration

3 months of treatment

Subjects

145 men and women aged 15-48 years, suffering from juvenile moderate polymorphic acne

Regimen

Applied Avène Diacnéal with fingers onto the face (except eyelids and mucous membranes) every evening after cleansing, without any rinsing or other cosmetic application within 4 hours after application

Methods of Assessment

Global scarring score

Number of inflammatory and retentional acne lesions on the face

Patients' self-assessments of global efficacy

Comparison of standardized digital photographs pre- and post-treatment

Side Effects

Burning sensation and erythema (redness of the skin), which caused 7 subjects in the Avène Diacnéal group and 2 subjects in the vehicle group to stop treatment

Effects observed in this study:

The numbers of acne lesions and comedones significantly decreased in subjects using Avène Diacnéal starting from day 28. Subjects using vehicle cream experienced a decline in the total number of comedones, but not in the number of inflammatory acne lesions. Avène Diacnéal was also more efficient than the vehicle on scarring in compliant patients, due to improvements in erythema and hyperpigmentation.

Interest of the association retinaldehyde/glycolic acid in adult acne.

Trial Design

Open-label, multi-center, uncontrolled

Duration

90 days of treatment

Subjects

397 women with acne who were aged between 30-40 years. Mainly of Fitzpatrick phototypes II, III and IV, and normal, oily or combination skin.

Regimen

Applied Avène Diacnéal in the evening alone or in association with other topical or systemic acne treatments.

Methods of Assessment

Counts of inflammatory lesions (papules and pustules) and retentional lesions (open and closed comedos).

Overall efficacy assessed by investigators using a 5-grade rating scale.

Side Effects

96.7% of the subjects did not experience any side effects. Adverse events were infrequent and included irritation, tightening and itching.

Conflicts of Interest

Although no conflicts of interest were declared, 3 out of 9 authors appeared to be employees of Avène.

Effects observed in this study:

Subjects who used Avène Diacnéal in combination with other systemic and oral anti-acne treatments had significantly lower numbers of inflammatory and retentional acne lesions. Specifically inflammatory lesion count declined by 46% between day 0-30 and by 50% between day 30-90, while retentional lesion count declined by 30% from day 0-30 and 44% from day 30-90. Overall efficacy was good or very good for 93% of the patients.

Subjects who used Avène Diacnéal alone without any other acne treatment also experienced significant reductions in inflammatory and retentional lesions, with 56% and 57% decreases in the counts of inflammatory and retentional lesions after 30 and 90 days of treatment, respectively. Overall efficacy was reported to be good or very good for 88% of the patients.

Trial Design

Multicenter

Duration

90 days of treatment

Subjects

1709 men and women aged 15-40 years, with acne

Regimen

Applied Avène Diacnéal once a day on the face, with or without other topical or systemic anti-acne drugs

Counts of the number of acne lesions on the face, and investigator evaluations of global efficacy using a 5-point scale

Side Effects

91% of subjects did not encounter any adverse events. Most side effects were noticed just after application, and lasted less than a few hours.

Effects observed in this study:

Inflammatory and retentional lesions both declined significantly after 30 and 90 days of treatment. Global efficacy was reported to be good or very good for 61% of the subjects overall, and for 59% of the subjects who used Avène Diacnéal alone without any other anti-acne treatments.
