Methods of Assessment

Clinical assessments of fine lines and wrinkles, dyspigmentation, overall grade of photoaging, scored according to the Griffiths photonumeric scale for photoaging.

Clinical assessment of tactile roughness, scored on a similar 0-8 scale.

Evaluation of skin biopsies from 28 participants for the expression of fibrillin-1 in the papillary dermis.

Effects observed in this study:

After 6 months, the serum led to a significant improvement in facial wrinkles over baseline, while the vehicle did not. This improvement was not statistically significant compared to that produced by the vehicle, but became statistically significant over that projected for the vehicle after an additional 6 months' of use.

Skin texture was also improved relative to baseline, but was not significantly better than the improvement seen with the vehicle. Neither the serum nor its vehicle provided any benefit to the appearance of mottled dyspigmentation or actinic lentignes.

The biopsies revealed a significant accumulation of fibrillin-1 in the papillary dermis of the skin after 6 months, which was significant compared to the vehicle. This restoration of fibrillin-1 is notable because fibrillin-rich microfibrils are key structural components of the elastic fiber network and are known to be reduced in photoaged skin.
