Effect of a novel dietary supplement on skin aging in post-menopausal women.

Subjects

100 women aged 45-65 years who were of Fitzpatrick phototypes II or III, were 1-5 years postmenopausal and had mild to moderate photodamage to the back of the hands.

Regimen

All subjects replaced their usual moisturizer and cleanser with the supplied products (Purpose Liquid Cleansing Wash and Purpose Dual Treatment Moisture Lotion SPF 15) during a 28-day washout period and for the duration of the study.

50 subjects were randomized to treatment with Imedeen Prime Renewal and 50 to treatment with the placebo. Subjects in both groups took 2 tablets in the morning and another 2 in the evening.

Methods of Assessment

Clinical grading of the face, décolletage and hand by one examiner, on a 10-point scale.

Evaluations of digital photographs of the full face and the full face plus décolletage using the same 10-point scale.

Ultrasound measurements of the combined epidermal and dermal thickness.

Measurement of skin moisturization using a NOVA Dermal Phase Meter.

Measurement of skin elasticity with a cutometer.

Subject self-assessment questionnaire

Side Effects

One participant in the active group withdrew from the study due to an adverse event (swollen tongue).

Effects observed in this study:

After 6 months, the clinical grades for forehead wrinkles, periocular wrinkles, perioral wrinkles, mottled hyperpigmentation, skin laxity, skin sagging, dark under-eye circles and overall appearance were all significantly improved in the subjects taking Imedeen Prime Renewal compared to the subjects on the placebo. Tactile roughness and telangiectasia were the only clinical parameters that were not significantly improved relative to the placebo group.

The evaluations of the digital photographs likewise found significant differences between the 2 groups for forehead wrinkles, periocular wrinkles, mottled hyperpigmentation and overall appearance. Perioral fine wrinkles, telangiectasia and dark under-eye circles were not significantly better compared to the improvements attained with the placebo after 6 months, however.

Clinical grading also showed significant differences in favour of the active treatment for décolletage crepiness, overall appearance of the décolletage, hand crepiness and mottled hyperpigmentation of the hand by the end of the treatment. The ultrasound measurements revealed that the dermal density at the crow's feet area was also significantly different between the 2 treatment groups. Finally, skin hydration and elasticity were not statistically different between the active and placebo-treated groups.

Marine-derived nutrient improves epidermal and dermal structure and prolongs the life span of reconstructed human skin equivalents.

Results / Report

The Marine Complex stimulated the expression and deposition of type I collagen when added to cultures of human skin equivalents (HSEs). It also reactivated epidermal proliferation of "mature" HSEs, increasing the number of viable cell layers and improving epidermal morphology. Moreover, it did so without affecting cellular differentiation or inducing epidermal stress. Finally, it had a remarkable effect on the deposition of the basement membrane components laminin 332 and collagen type VII at the dermal-epidermal junction.

Most of the effects were more pronounced in female HSEs than in male HSEs.

Supplementation with a complex of active nutrients improved dermal and epidermal characteristics in skin equivalents generated from fibroblasts from young or aged donors.

Results / Report

In vitro reconstructed skin equivalents (SEs) supplemented with the complex of nutrients found in Imedeen Prime Renewal led to profound effects on epidermal and dermal morphology. It increased the density of the extracellular matrix and resulted in a thicker and better organized epidermal structure, particularly in the the SEs generated with fibroblasts from the 49-year-old donor (as compared to those from a 19-year-old donor). It also seemed to delay the onset of senescence.

In addition, the treatment had differing effects on collagen I and fibrillin I mRNA expression depending on the age of the donor fibroblasts used to generate the SEs. It significantly enhanced the expression and deposition of type 1 collagen and fibrillin-1 in the SEs generated with the older fibroblasts, but not those generated with the younger fibroblasts. Elastin expression was not significantly affected even in SEs generated from the older fibroblasts, but there was a positive tendency.

Finally, the epidermis of the SEs that had received supplementation had a greater number of keratinocyte layers, probably due to its stimulation of cell proliferation.
