Subjects

41 women with moderate photoaging (dyschromic facial skin with fine lines and wrinkles) between the ages of 30 and 65.

Regimen

Applied either the 1% idebenone lotion (Prevage MD) or the 0.5% idebenone lotion (Prevage) to the entire face twice daily, once in the morning and once in the evening.

Methods of Assessment

Electrical conductance measurements of skin surface hydration

Expert grader assessment of improvements of skin roughness/dryness, fine lines/wrinkles, and overall global improvement on a 5-point scale, using high-resolution, standardized digital photographs taken at baseline, 3 weeks and 6 weeks.

Patient diaries and questionnaires to gauge perceived experiences with the product.

Effects observed in this study:

Prevage MD: After 6 weeks of use of the 1% idebenone formulation, the women achieved a 26% decline in skin roughness/dryness, a 29% decline in fine lines/wrinkles, a 37% increase in skin hydration and an overall 33% improvement in the global assessment of photodamaged skin.

Prevage: At the end of the study, the subjects who had used the 0.5% idebenone lotion observed a 23% reduction in skin roughness/dryness, a 27% reduction in fine lines/wrinkles, a 37% increase in skin hydration and an overall 30% improvement in the global assessment of photodamaged skin.

Both products: Comparison of the digital photographs showed that there were visible improvements in periorbital rhytides and pigment dyschromia for both groups of subjects. Histological analysis revealed a general improvement in the epidermis, an increase in dermal collagen content, and decreases in matrix metalloproteinase-1, IL-6 and IL-1b.

A 50 year old woman developed a facial rash after using the antioxidant facial cream Prevage. 4 hours after application, her face felt hot and tight, and by the following morning her upper chest was red and swollen and she had swelling around her eyes. The irritation lasted for 2 weeks, settling with dryness and scaling.

Patch testing of the ingredients at the concentrations present in the product revealed a positive reaction to 0.5% idebenone.

Allergic contact dermatitis to idebenone in an over-the-counter anti-ageing cream.

A 43 year old Caucasian woman developed an itchy rash within 2 days of applying Prevage anti-aging cream to her face and neck. The rash was resolved with the use of a topical steroid.

Patch testing of her cosmetics showed a positive reaction to the Prevage cream, specifically the 0.5% idebenone. She also had a positive reaction to nickel.
