Introduction

What is estrone?

Estrone is a steroid hormone that is a precursor of estradiol, another naturally occurring estrogen in humans (R). It is weaker estrogen than estradiol, but stronger than estriol (R, R, R). Its production in women increases with age and excess body weight, with morbidly obese postmenopausal women producing as much as 10x more estrone (R, R, R).

Is estrone stable?

Formulations containing 1% estrone in addition to 1% estradiol, 2% estriol, 20% progesterone and 5% testosterone have been demonstrated to remain stable when incorporated in 3 different cream bases and refrigerated or stored at room temperature for 6 months (R).

Does estrone get absorbed?

Estrone has much higher permeability and diffusion constants than both estradiol and estriol, indicating that it is absorbed well by the skin by diffusing through the stratum corneum (R).

What happens after estrone is absorbed?

When human skin was incubated with estrone, only about 3% of the estrone was converted to estradiol (R, R).
